Nanocarriers Tryst with the Lysosomes: Implications in Intracellular Drug Delivery and Disease

The deployment of nanoparticles that can deliver drugs/genes inside cells by exploiting endocytosis has shown tremendous potential for biology and medicine. Yet, almost all nanoparticles once captured by the cell are routed primarily towards the lysosomes. My talk will focus on 1. Dissection of intracellular transport of nanoparticles for mRNA delivery using cells that are genetically edited for select endosomal proteins and identification of novel compounds that improves cytosolic delivery. 2. We have utilized the inherent tropism of nanocarriers towards the lysosomes for delivery of therapeutics for the delivery of drugs to these vesicles for treatment of Niemann Pick Type C-1, a lysosomal storage disorder (LSD). These studies will provide insights into development of novel nanotechnologies for the treatment of wide variety of LSDs.
